POU1F1 is a transcription factor that can push breast cancer cells toward a cancer stem cell‑like state. When POU1F1 levels are high, the cells produce more IL‑6, which activates the JAK2/STAT3 signaling pathway. This pathway is known for supporting cell survival, growth, and stem‑like behavior, and its activation leads to increased ALDH expression, a marker commonly associated with cancer stem cells. Through this mechanism, breast cancer cells gain several aggressive traits: they form more mammospheres, show stronger clonogenic growth, become more invasive, shift their metabolism toward glycolysis, and develop resistance to common cancer treatments. Tumors with high POU1F1 expression, especially certain luminal breast cancers, may therefore be more difficult to treat because they contain a larger population of stem‑like cells. This makes the POU1F1–IL‑6/JAK2/STAT3–ALDH axis an important area of interest for developing therapies that target cancer stemness and improve treatment outcomes.
